BASEBALL

ESU BASEBALL WINTER CLINIC

The ESU Baseball program would like to invite T-ball, Little League, middle school and high school players to attend a developmental clinic series with college-level instruction in hitting, defense, and pitching. Players will participate in their age group. The clinic will be held at Koehler Fieldhouse, cost is $50 per session. Attend all six sessions get one free ($250). The sessions are as follows:

Prepare for the upcoming season. The hitting clinic consists of swing mechanics, station work, and live hitting. Defensive clinic consists of positional group work with pitchers and catchers working on proper mechanics and techniques.

TITANS BEGIN WINTER WORKOUTS

The NorthEast Titans baseball club will soon start its annual winter workouts and conduct tryouts for its travel baseball teams (age as of April 30, 2013). The workouts will incorporate proven methods that emphasize proper mechanics and baseball fundamentals. If you are interested in travel baseball and/or you want to receive quality offseason instruction from experienced coaches, please contact Blaine Fardella at b.fardella1@gmail.com for the 10U level or Kevin Dillon (northeasttitansbaseball@gmail.com) at the 8U and 9U levels. With the recent arrival of winter, now is the perfect time to hone and refine your essential fly-tying skills. In its continuing effort to provide free fly-tying classes and workshops to the general public, as well as its own members, the Brodhead Chapter of Trout Unlimited is offering a unique Saturday morning program to kick off its winter fly-tying programs and activities. On three consecutive Saturdays, Jan. 5, 12 and 19 from 9 am until 12 p.m., members of BTU will be on-hand at the Monroe County Environmental Education Center located at 8050 Running Valley Road in Bartonsville, right off Route 611, to teach their favorite and highly effective Pocono trout-catching patterns! The chief instructor for this course is PA State Champion fly-tier Scott Cesari, and he will be joined by the Master Fly-Tiers of the Brodhead TU Chapter. Bring your vise and fly-tying tools and learn how to tie flies that really catch trout from some of the best fly-tiers in the northeast, including Eric Baird, Tom Battista, Don Baylor, Gerry Bortz, Todd Burns, Will Daskal and Bob Stevens. As always, instruction for this comprehensive course is absolutely free; however, there is a small $15 fee that simply covers the material packets required to tie several flies for each of the patterns covered in class.

NEW CHALLENGER SOCCER DIVISION

Our new Challenger Soccer division will be presented through our partnerships with the Pocono Family YMCA and the Pocono Mountain School District and join the current offerings of baseball and basketball. These programs are for youth who are developmentally and/or physically challenged. Our newest offering is designed to help players to enjoy the sport of soccer in a fun, safe and non-competitive environment. Challenger sports opportunities began six years ago with the baseball team sponsored through our partnership with the Big Pocono Little League and has had terrific response. With our partnership organizations and sponsorships the Challenger crew is able to provide the opportunities for all kids to be participants, rather than spectators cheering for their siblings. In order to be able to move forward, we need the support of local companies for sponsorships of the four new soccer teams and donations from the community to help purchase equipment for the teams. Our goal is $1,500 of which $1,000 is via sponsorships. We have already received a commitment for one team sponsorship. Our season begins on Sunday, Oct. 6 and runs through Sunday, Nov. 11. The staff of ESU track & field will be hosting its 5th annual Holiday Field Clinic at Koehler Fieldhouse Thursday, Friday and Saturday, Dec. 27-29 from 9 a.m.-4 p.m. These are for field events ? Shot, Discus, Javelin, Long Jump, Triple Jump, High Jump. Learn the techniques and drills we teach our Warrior athletes to get ready for your upcoming season. Gunman dead after shooting 3 cops at station

Three New Jersey police officers are in the hospital after being shot inside a police station by a man being processed for a domestic incident. MSNBC's Thomas Roberts reports.

The gunman who shot three officers after a "violent struggle" at a New Jersey police station early Friday is dead, officials said.

The suspected shooter had been brought into Gloucester Township Police station?for a domestic incident when he got into a confrontation with officers, reported?NBC's Philadelphia affiliate, NBC 10.


"A violent struggle occurred while the suspect was being processed," Deputy Chief David Harkins said. The man was able to grab a gun and then opened fire.

One of the injured officers, who was rushed to surgery after being shot below his bulletproof vest, is listed in stable condition. The other two were treated for graze wounds and released.?

The shooter was killed by police during the confrontation, according to Chief W. Harry Earle.?The investigation was ongoing.?

Gloucester is located in central New Jersey, about 15 miles outside Philadelphia.

Baltimore's Washington Monument Damaged When Dozing Driver Crashes Into Wall

A car crashed into a Washington Monument park wall on Thursday at approximately 10 a.m., overturning and injuring a passenger and damaging the historic monument.

The flipped over four-door gray sedan blocked traffic on Washington Place just south of the 178-foot column to West Centre Street for at least an hour while police investigated and roped off a damaged stone wall in the park's South Garden.

Two men were in the car, which was a rental vehicle licensed in Connecticut, when the driver fell asleep and crashed into the wall, causing the car to flip over, police spokesman Det. Vernon Davis said. The driver was uninjured but paramedics transported the passenger to University of Maryland Medical Center with neck and back injuries.

Strange behavior: New study exposes living cells to synthetic protein

Dec. 27, 2012 ? One approach to understanding components in living organisms is to attempt to create them artificially, using principles of chemistry, engineering and genetics. A suite of powerful techniques -- collectively referred to as synthetic biology -- have been used to produce self-replicating molecules, artificial pathways in living systems and organisms bearing synthetic genomes.

In a new twist, John Chaput, a researcher at Arizona State University's Biodesign Institute and colleagues at the Department of Pharmacology, Midwestern University, Glendale, AZ have fabricated an artificial protein in the laboratory and examined the surprising ways living cells respond to it.

"If you take a protein that was created in a test tube and put it inside a cell, does it still function," Chaput asks. "Does the cell recognize it? Does the cell just chew it up and spit it out?" This unexplored area represents a new domain for synthetic biology and may ultimately lead to the development of novel therapeutic agents.

The research results, reported in the advanced online edition of the journal ACS Chemical Biology, describe a peculiar set of adaptations exhibited by Escherichia coli bacterial cells exposed to a synthetic protein, dubbed DX. Inside the cell, DX proteins bind with molecules of ATP, the energy source required by all biological entities.

"ATP is the energy currency of life," Chaput says. The phosphodiester bonds of ATP contain the energy necessary to drive reactions in living systems, giving up their stored energy when these bonds are chemically cleaved. The depletion of available intracellular ATP by DX binding disrupts normal metabolic activity in the cells, preventing them from dividing, (though they continue to grow).

After exposure to DX, the normally spherical E. coli bacteria develop into elongated filaments. Within the filamentous bacteria, dense intracellular lipid structures act to partition the cell at regular intervals along its length. These unusual structures, which the authors call endoliposomes, are an unprecedented phenomenon in such cells.

"Somewhere along the line of this filamentation, other processes begin to happen that we haven't fully understood at the genetic level, but we can see the results phenotypically," Chaput says. "These dense lipid structures are forming at very regular regions along the filamented cell and it looks like it could be a defense mechanism, allowing the cell to compartmentalize itself." This peculiar adaptation has never been observed in bacterial cells and appears unique for a single-celled organism.

Producing a synthetic protein like DX, which can mimic the elaborate folding characteristics of naturally occurring proteins and bind with a key metabolite like ATP is no easy task. As Chaput explains, a clever strategy known as mRNA display was used to produce, fine-tune and amplify synthetic proteins capable of binding ATP with high affinity and specificity, much as a naturally occurring ATP-binding protein would.

First, large libraries of random sequence peptides are formed from the four nucleic acids making up DNA, with each strand measuring around 80 nucleotides in length. These sequences are then transcribed into RNA with the help of an enzyme -- RNA polymerase. If a natural ribosome is then introduced, it attaches to the strand and reads the random sequence RNA as though it was a naturally-occurring RNA, generating a synthetic protein as it migrates along the strand. In this way, synthetic proteins based on random RNA sequences can be generated.

Exposing the batch of synthetic proteins to the target molecule and extracting those that bind can then select for ATP-binding proteins. But as Chaput explains, there's a problem: "The big question is how do you recover that genetic information? You can't reverse transcribe a protein back into DNA. You can't PCR amplify a protein. So we have to do all these molecular biology tricks."

The main trick involves an earlier step in the process. A molecular linker is chemically attached to the RNA templates, such that each RNA strand forms a bond with its newly translated protein. The mRNA-protein hybrids are exposed to selection targets (like ATP) over consecutive rounds of increasing stringency. After each round of selection, those library members that remain bound to the target are reverse-transcribed into cDNA (using their conveniently attached RNA messages), and then PCR amplified.

In the current study, E. coli cells exposed to DX transitioned into a filamentous form, which can occur naturally when such cells are subject to conditions of stress. The cells display low metabolic activity and limited cell division, presumably owing to their ATP-starved condition.

The study also examined the ability of E. coli to recover following DX exposure. The cells were found to enter a quiescent state known as viable but non-culturable (VBNC), meaning that they survived ATP sequestration and returned to their non-filamentous state after 48 hours, but lost their reproductive capacity. Further, this condition was difficult to reverse and seems to involve a fundamental reprogramming of the cell.

In an additional response to DX, the filamentous cells form previously undocumented structures, which the authors refer to as endoliposomes. These dense lipid concentrations, spanning the full width of the filamented E. coli, segment the cells into distinct compartments, giving the cells a stringbean-like appearance under the microscope.

The authors speculate that this adaptation may be an effort to maintain homeostasis in regions of the filamentous cell, which have essentially been walled off from the intrusion of ATP-depleting DX. They liken endoliposomes to the series of water-tight compartments found in submarines which are used to isolate damaged sections of the ship and speculate that DX-exposed cells are partitioning their genetic information into regions where it can be safely quarantined. Such self-compartmentalization is known to occur in some eukaryotic cells, but has not been previously observed in prokaryotes like E. coli.

The research indicates that there is still a great deal to learn about bacterial behavior and the repertoire of responses available when such cells encounter novel situations, such as an unfamiliar, synthetic protein. The study also notes that many infectious agents rely on a dormant state, (similar to the VBNC condition observed in the DX-exposed E. coli), to elude detection by antibiotics. A better understanding of the mechanisms driving this behavior could provide a new approach to targeting such pathogens.

The relative safety of E. coli as a model organism for study may provide a fruitful tool for more in-depth investigation of VBNC states in pathogenic organisms. Further, given ATP's central importance for living organisms, its suppression may provide another avenue for combating disease. One example would be an engineered bacteriophage capable of delivering DX genes to pathogenic organisms.

How is hearing loss tested? - Ask Doctor K, Harvard Medical School

DEAR DOCTOR K:

I?m losing my hearing so my doctor has scheduled an audiological evaluation. What will this evaluation tell me?

DEAR READER:

An audiological evaluation identifies the severity of your hearing loss and the frequency range in which it occurs. This information may help identify the cause of your hearing loss. It?s also essential for determining whether you could benefit from a hearing aid and, if so, which style and type would help the most.

When a sound wave enters your ear, it hits the eardrum. Several tiny bones transmit sound waves from the eardrum through fluid, into the inner ear. There lies a snail-shaped organ called the cochlea that is covered with tiny hairs. These hairs generate electrical signals that travel along the hearing nerve (the auditory nerve) to the brain. Sound travels in waves through the air (A) into the ear canal (B), where it is amplified and vibrates the eardrum (C). The eardrum vibrates the tiny bones known as the ossicles (D), which transmit the vibrations through the oval window and into the fluid in the cochlea in the inner ear.

The cochlear fluid (E) moves in waves, setting in motion the sensory hair cells inside the organ. The hair cells release electrochemical signals that travel along more than 30,000 auditory nerve pathways into the brain (F).

The evaluation is done by a hearing specialist, or audiologist, in a specially constructed booth that shuts out all unwanted noise. It tests the different parts of the ear that are necessary for hearing:

  • Pure-tone test. Each of your ears, in turn, is exposed to sounds of different frequencies and decibel levels. (Frequency is the pitch of a sound. A decibel is the measurement of the loudness of a sound.) The pure-tone test identifies the quietest tones that you can hear at different frequencies.
  • Next is a bone-conduction test. This test helps establish whether your hearing loss is primarily sensorineural (caused by damage to the cochlea, hair cells or auditory nerve) or conductive (caused by a blockage of sound transmission through your outer or middle ear), or a combination of the two. This test will also help determine whether your hearing loss can be medically corrected, and the kind of hearing aid you need.

Two additional tests will evaluate how well you hear and understand spoken words:

  • Speech reception threshold test. You will hear words, instead of tones, through your earphones. The words start out loud and gradually get softer. The test identifies the decibel level at which you can understand and repeat only half of the words.
  • Speech discrimination test. With age, many people can hear words loudly but not clearly. A speech discrimination test assesses how well you understand words. A high score on this test means that you are likely to benefit from a hearing aid. That?s because your problem in understanding a spoken word comes from not hearing the word loudly enough, and a hearing aid can amplify the loudness of sounds.

Today?s technology for testing your hearing has improved greatly since I was in medical school. Even greater advances have been made in hearing aids. They not only work better but they?ve also become tiny ? people often don?t notice you have one. For many of my patients, hearing tests followed by the right hearing aid has greatly improved the quality of their lives.

Investing in big data's second act - Fortune Finance: Hedge Funds ...

The list of life's certainties has gotten longer. Along with death and taxes we can now include information overload. The amount of data generated on a daily basis has exploded, overwhelming people and organizations alike. This creates an opportunity for many types of investors, including private equity firms, to turn information overload into smart data that can drive decision-making at the highest levels of enterprise.

For private equity investors this means two things: Investing in the emerging leaders who are building the apps, infrastructure and services that businesses need in order to capitalize on the data revolution, and ensuring that our portfolio companies are effectively using these capabilities to drive the success of their businesses.

Welcome to the Big Data opportunity:? Research firm IDC expects Big Data to grow from $3.2 billion in 2010 to $16.9 billion in 2015. Peter Sondergaard, Gartner's global head of research, says that by 2015 we'll see 4.4 million jobs devoted to the global support of Big Data, with 1.9 million based in the U.S. He also predicts a jobs multiplier effect ? each IT job created by Big Data will generate three more positions outside of IT.

How new is this era, exactly? Some say that we are still in the nascent stages of the Big Data revolution, and the way to participate is via early-stage venture capital firms taking stakes in young startups. This is actually not the case. What we are witnessing is the evolution of enterprise intelligence, with companies continuing to refine their data strategies and sector winners beating their competitors by using their customer, market and product data more effectively.

Until now, Big Data was not synonymous with "smart data." Business intelligence until recently has meant enabling IT departments to warehouse and organize information that might be reviewed periodically by senior managers.? The intelligence was on the periphery of the decision-making process. Still, most enterprises adopted these tools very broadly and investors in these companies profited. IBM acquired Cognos.? Oracle acquired Hyperion.? And innovation moved on to the next set of emerging leaders.

The second act of Big Data is about action. This time intelligence will go beyond reporting. It will live at the heart of the organization, not at the edges. It will involve moving from historical insight to predictive information, enabling managers to make smarter decisions. To match these new capabilities, enterprises are building a new layer of infrastructure, applications and services.? No industry will remain untouched.? Investors have many options for participating in Big Data. While winners will be found among the startups and early stage venture capital firms providing the infrastructure layer, we believe that big successes will come from enabling the business users, providing intelligence applications and services or designing entirely new business models within verticals.

The winners of the second act of Big Data are now emerging. How can an organization place itself among them? First, it needs to understand the opportunities and the threats of this continuing, massive change. Second, it needs to start investing now in people and technology in order to avoid missing out as the second act unfolds.

A new set of Big Data investment opportunities exists in players ranging from the law enforcement and healthcare sectors to finance and government. We believe that Big Data is transforming how enterprises approach business issues. Providers of enabling technologies and business innovators all have opportunities for enormous success here. Who will be the winners in Big Data's second act? The list will certainly include private equity firms who can effectively direct investment in the trend. As for generalist investors, they should make sure their portfolio companies have an eye on the Big Data evolution to leverage their mountains of data and to see the road ahead.
